What Poison Cloud does

The skill text is one sentence: releases poison, dealing damage to surrounding enemies. Two things follow from the word surrounding.

First, it does not care which way Poison Mushroom is facing. A slash or a shot only hits what is in front of it, so it can miss when enemies flank or when the frontline shifts. A cloud centred on the caster hits whatever is close, in any direction. On a Charge Cookie, the class that runs into the middle of things, being surrounded is the normal state rather than the failure state.

Second, the damage is spread rather than concentrated. There is nothing here that focuses a single target, and nothing that gathers, pushes, stuns or shields. It is area damage and nothing else.

The published data does not include per-stat base values for Poison Mushroom, so we are not going to guess at ATK, DEF or HP. The stat templates that are documented cover SSR Cookies only; no clean R Charge template has been published, and inventing one would be worse than leaving the gap visible.

Range Synergy: four providers, and one is U rarity

Cream Soda Cookie is the best pairing, on cooldown alone. Synergy effects are tied to the provider’s skill firing, so a 3.5-second provider applies its buff far more often than a 7.0-second one. Cream Soda is also a Charge, which means it is standing in roughly the same place Poison Mushroom is.

The more useful entry for a new account is Knight Cookie. Knight is U rarity, the cheapest tier in the game, and still cycles at 5.0 seconds, faster than both the SR Macaron and the SSR Toothpaste. This is the pattern the Synergy system keeps repeating: the buff you need is often held by a Cookie you already own and were not paying attention to. A player with no SSRs at all can run Poison Mushroom with their Range Synergy live, using a U-rarity Tank they were probably fielding anyway.

Range increases how far a skill reaches. On a cloud that already covers the area around the caster, a wider radius means more enemies caught per cast, which for a pure-damage area skill is close to a straight damage multiplier. Full provider lists for every Synergy are on the Synergy page.

It does not grant a Synergy

Only 18 of the 69 launch Cookies grant a Synergy to their team. Poison Mushroom is not one of them. That matters more than it sounds, because granting a Synergy is the one thing that keeps a low-rarity Cookie on a roster permanently. Nobody benches the U-rarity Knight, because Knight is one of only four sources of Range in the entire game.

Poison Mushroom has no such job. They receive a buff and give nothing back, so their slot is competed for purely on the strength of Poison Cloud.

Competition: the slowest of the four R Charge Cookies

There are four R Charge Cookies, and the cooldown spread between them is wide.

Poison Mushroom is last on this list, and not by a small margin. Bear Jelly Worker fires more than three times for every one Poison Cloud. That is the honest headline and it should not be dressed up.

The counter-argument is supply, not power. Bear Jelly Worker and Dark Cherry both depend on Rapid Fire, which only two Cookies in the game grant. Poison Mushroom’s Range has four providers including a U-rarity one. A player who owns neither Rapid Fire source ends up with a buffed Poison Mushroom and an unbuffed Bear Jelly Worker, and that comparison is much closer than the raw cooldowns suggest.

That is a real argument, but it is an argument about your account, not about the Cookie. Once a Rapid Fire provider shows up, the faster Charges pull ahead again.

Team fit

Because they receive Range, Poison Mushroom shares providers with several other Cookies, so a Range source you are already running for someone else covers them at no extra cost. Cream Soda and Toothpaste in particular tend to be on teams for their own sake.

As a Charge they belong in the front half of the formation, which suits a skill that damages everything nearby. Pair them with something that holds enemies in place. The longer a group stays clustered, the more of them a 5.75-second cloud catches.

Should you build them?

Early on, yes. Poison Mushroom is easy to use, needs no aiming, and their Synergy is one of the least demanding in the game to satisfy. If you have Knight Cookie, you have a working Poison Mushroom.

Later, replace them. They do not grant a Synergy, they have no utility beyond damage, and three same-class, same-rarity Cookies cycle faster. There is no version of a finished roster where the 5.75-second option beats the 1.75-second one at the same job.

Keep them longer than that in one case only: you have no Rapid Fire provider and no Projectile Speed provider, and Range is the only Synergy you can actually turn on. In that situation a buffed Poison Mushroom really is the better Charge, and it is worth checking your provider list before writing them off.
